Molecular Activation Mechanism and Structural Dynamics of Orange Carotenoid Protein

چکیده
Like most photosynthetic organisms, cyanobacteria are vulnerable to fluctuations in light intensity, which can damage their machinery. To protect against this, they use a photoprotective mechanism called non-photochemical quenching (NPQ), where excess absorbed photo-energy is dissipated as heat. In cyanobacteria, activation of Orange Carotenoid Protein (OCP) the critical first step NPQ response. OCP also only known photosensitive protein, uses carotenoid for its activation. We summarize current knowledge on induced reactions OCP; different mechanisms that have been proposed; photocycle kinetics and characteristics; reported structural intermediates. discuss possible interpretations experimental results, we formulate important open questions directions future work, reveal molecular basis photosensing by OCP.

The Signaling State of Orange Carotenoid Protein.
Orange carotenoid protein (OCP) is the photoactive protein that is responsible for high light tolerance in cyanobacteria. We studied the kinetics of the OCP photocycle by monitoring changes in its absorption spectrum, intrinsic fluorescence, and fluorescence of the Nile red dye bound to OCP. It was demonstrated that all of these three methods provide the same kinetic parameters of the photocycl...

Photoprotective mechanisms are of fundamental importance for the survival of photosynthetic organisms. In cyanobacteria, the orange carotenoid protein (OCP), when activated by intense blue light, binds to the light-harvesting antenna and triggers the dissipation of excess captured light energy. Using a combination of small angle X-ray scattering (SAXS), X-ray hydroxyl radical footprinting, circ...
متن کاملStructural determinants underlying photoprotection in the photoactive orange carotenoid protein of cyanobacteria.
The photoprotective processes of photosynthetic organisms involve the dissipation of excess absorbed light energy as heat. Photoprotection in cyanobacteria is mechanistically distinct from that in plants; it involves the orange carotenoid protein (OCP), a water-soluble protein containing a single carotenoid. The OCP is a new member of the family of blue light-photoactive proteins; blue-green li...
متن کاملOrange Carotenoid Protein Quenches Excess Energy and Singlet Oxygen.
Harnessing light for energy gives photosynthetic organisms free energy—great, right? Well, like so many free things, too much of a good thing can turn into a bad thing, like a drought-ending rain that turns into a levybreaching flood. The photosynthetic apparatus, beautifully adapted to capture light, can incur serious damage when too much light energy floods in. Given the variability of light ...

β-Carotene adds nutritious value and determines the color of many fruits, including melon (Cucumis melo). In melon mesocarp, β-carotene accumulation is governed by the Orange gene (CmOr) golden single-nucleotide polymorphism (SNP) through a yet to be discovered mechanism. In Arabidopsis (Arabidopsis thaliana), OR increases carotenoid levels by posttranscriptionally regulating phytoene synthase ...
